When you click on your backpack, look in the upper left hand corner of your inventory screen. Numbers there indicate how many things you have in your backpack/how many things you can carry. Next to this is a white + in a yellow circle. By clicking this you get the option to purchase additional slots in your inventory with $ or skulls (depending, I believe, on your level).
