If I choose the option to start over, do I lose more than my fans? Do I get to keep my businesses and homes?

If you mean the Comeback, all you lose are fans. If you start your whole came over, you go back to nothing, you customize your doll from scratch, with nothing other than the basics that you start out with. Now, if it's the Comeback, then you increase your maximum energy by 3, you get an outfit, keep all your stuff, you only lose fans and plummet to E+ Lister. Which are you talking about?

I was wondering, what happens to the missions? Do you have to repeat all the beginning missions or do you continue from where you were in the storyline when you took the comeback?

No, you don't go back and redo the original tasks and gigs and storylines. You just keep moving along where you were, from a much lower rank.
